本研究探讨基因IEX-1启动子区CpG岛甲基化状态的改变及其与恶性血液病发生的相关性。应用MSP的方法检测9种恶性血液病细胞系中IEX-1基因启动子区CpG岛甲基化状态,并把经M.sssI酶处理过的正常人外周血单个核细胞中IEX-1 CpG岛甲基化状态作为阳性对照,把正常人外周血单个核细胞中IEX-1 CpG岛甲基化状态作为阴性对照。结果显示:在NB4、Molt4.、Raji细胞系中IEX-1基因启动子区CpG岛呈高甲基化状态;在CA46、CEM、U937、K562、HL-60、Jurkat细胞系中IEX-1基因启动子区CpG岛呈部分甲基化状态;在正常人外周血单个核细胞中IEX-1基因启动子区CpG岛呈非甲基化状态;经M.sssI酶处理过的正常人外周血单个核细胞中IEX-1基因启动子CpG岛呈高甲基化状态。结论:IEX-1基因启动子区CpG岛甲基化状态的改变与恶性血液病有一定的相关性。
This study was aimed to investigate the change of methylation status of CpG island in gene promoter region IEX-1 and its relationship with the occurrence of hematologic malignancies. MSP method was used to detect the methylation status of CpG island in promoter region of IEX-1 gene in 9 hematological malignant cell lines and the IEX-1 CpG island in normal human peripheral blood mononuclear cells treated with M.sssI Methylation status as a positive control, the normal human peripheral blood mononuclear cells IEX-1 CpG island methylation status as a negative control. The results showed that the CpG island of promoter region of IEX-1 gene was hypermethylated in NB4, Molt4 and Raji cell lines. The promoter of IEX-1 gene in CA46, CEM, U937, K562, HL-60 and Jurkat cell lines CpG island was partially methylated in normal human peripheral blood mononuclear cells; CpG island in promoter region of IEX-1 gene was non-methylated; normal human peripheral blood mononuclear cells treated with M.sssI The CpG island of IEX-1 promoter was hypermethylated. Conclusion: The change of methylation status of CpG island in promoter region of IEX-1 gene is associated with hematologic malignancies.
