目的:采用自制点状烫伤器建立小鼠双烫伤模型,比较干馏鸭蛋黄油和干馏鸡蛋黄油对烫伤的治疗效果。方法:取健康昆明种小鼠,雌雄各半,分为高体重鼠(33.44±3.97)g和低体重鼠(27.42±1.66)g。小鼠胶条固定,使用点状烫伤器在以腰椎为对称中心的两侧臀浅肌处体表烫出直径1.5 cm的圆形烫伤区,以小鼠初结痂时间和对结痂完全的皮肤组织学检查为指标,考察点状烫伤器接触时间、鼠重对造模情况的影响。小鼠深Ⅱ°烫伤造模后随机分为14个组,即模型对照组,磺胺嘧啶银治疗组为阳性对照组,鸡蛋黄油治疗组、鸭蛋黄油治疗组、鸡蛋黄油和鸭蛋黄油组合治疗组、鸡蛋黄油与磺胺嘧啶银组合治疗组、鸭蛋黄油与磺胺嘧啶银组合治疗组,每组均按雄雌分别编号。记录各组的烫伤创面愈合时间,考察不同药物的治疗效果。结果:5 s高体重鼠和低体重鼠烫伤组、10 s高体重鼠烫伤组可以完全成功造模,10 s低体重鼠烫伤组、25 s高体重鼠和低体重鼠烫伤组均死亡。鼠体重对初结痂时间和烫伤损伤程度有一定的影响;其中5 s烫伤组,高体重鼠与低体重鼠的初结痂时间有显著差异(P<0.01)。在雄鼠实验组,与模型对照组相比,阳性对照组可以显著缩短愈合时间(P<0.01),阳性对照组与蛋黄油组合治疗组就愈合时间无显著差异(P>0.05),鸭蛋黄油(P<0.05)促进愈合效果不及鸡蛋黄油(P<0.01)。对比雌雄两个实验组,雌鼠表现出更强的自愈能力(P<0.01),鸭蛋黄油促进创面愈合存在显著雌雄差异(P<0.01),而鸡蛋黄油无此类情况(P>0.05)。结论:所采用的点状烫伤器易于控制烫伤程度,易于复制较为稳定的皮肤烫伤模型;鸭蛋黄油有促进雄鼠创面愈合作用,但效果不及鸡蛋黄油治疗组。
OBJECTIVE: To establish a mouse model of double scald injury using homemade point-shaped scald device and compare the therapeutic effects of defatted duck egg butter and retted egg butter on scald. Methods: Healthy Kunming mice were divided into two groups, male and female mice. The mice were divided into three groups: high-weight mice (33.44 ± 3.97g) and low-weight mice (27.42 ± 1.66g). Mouse strip fixed, the use of point-like scald in the lumbar spine on both sides of the muscular symmetry center of the surface of the body scalded 1.5 cm in diameter circular scalding area, the initial mouse crusting time and the complete crusting Skin histological examination as an indicator to investigate the point-like scalder contact time, the impact of rat weight on the modeling. The mice were randomly divided into 14 groups, namely model control group, silver sulfadiazine treatment group, positive control group, egg-butter treatment group, duck-egg-butter treatment group, combination of egg-yolk and duck egg-butter treatment group, Egg butter and silver sulfadiazine combination therapy group, duck egg butter and silver sulfadiazine combination therapy group, each group were numbered according to male and female. Record the burn wound healing time of each group and examine the therapeutic effect of different drugs. Results: The 5-s high-body weight and low-body-weight scald group and the 10-s high body weight scald group were completely successful. The 10-s low-body weight scald group, 25-s high-body weight and low-body weight scald group all died. The body weight of rats had some influence on the time of initial scab and the degree of scald injury. The initial scab time of 5-s scald group, high body weight and low body weight rats were significantly different (P <0.01). Compared with the model control group, the positive control group can significantly shorten the healing time (P <0.01), and the positive control group and the egg yolk oil combination treatment group had no significant difference in the healing time (P> 0.05) (P <0.05) to promote healing less effective than egg butter (P <0.01). Compared with the control group, the female mice exhibited more self-healing ability (P <0.01), but there was significant difference between male and female in promoting the wound healing with duck egg butter (P <0.01) . Conclusion: The point scalder is easy to control the degree of scald, easy to copy a more stable model of skin scalds; duck egg butter can promote the wound healing of male rats, but the effect is not as good as egg butter treatment group.
