使用非抗原性聚乙二醇(PEG)修饰羊生长激素(OVine growth hormone,oGH),分别获得氨基修饰率为35%和86%的PEG-oGH。氨基修饰率为35%的PEG-oGH仍保留了与天然oGH抗血清的反应性,但修饰率为86%的PEG-oGH则丧失了。荧光光谱分析表明,与天然oGH比较,修饰率为35%的PEG-OGH仅其荧光强度有所减弱,光谱图并未改变,但修饰率为86%的PEG-oGH则发生了明显的图谱变化。体内结合试验表明,两种修饰oGH与肝受体结合之能力均有明显降低。
Ovine growth hormone (oGH) was modified with non-antigenic polyethylene glycol (PEG) to obtain PEG-oGH with 35% and 86% amino group modification respectively. PEG-oGH with a 35% amino group modification retains reactivity with the native oGH antiserum, whereas PEG-oGH with a modification rate of 86% is lost. Fluorescence spectrum analysis showed that the fluorescence intensity of PEG-OGH with a modification rate of 35% was weakened compared with that of the natural oGH, and the spectrum did not change, but the obvious change of the spectrum was observed when the modification rate of PEG-OGH was 86% . In vivo binding experiments showed that the two modified oGH and liver receptor binding capacity were significantly reduced.
