目的:探索肝细胞在肝纤维化形成中的可能作用。方法:用核酸原位分子杂交方法和3H脯氨酸掺入法,观察原代培养大鼠肝细胞在血小板衍生生长因子(PDGF)刺激前后胶原合成和Ⅰ、Ⅲ型前胶原mRNA的表达及钙拮抗剂维拉帕米对此作用的影响。结果:肝细胞培养一定时间后能够合成胶原,能够表达Ⅰ、Ⅲ型前胶原基因。PDGF能促进肝细胞的胶原合成和前胶原基因表达。但此刺激作用为维拉帕米所抑制。结论:这些研究为肝细胞参与肝纤维化形成提供了实验依据。
Objective: To explore the possible role of hepatocytes in the formation of hepatic fibrosis. Methods: The in situ hybridization of nucleic acids and incorporation of 3H-proline were used to observe the collagen synthesis and the expression of procollagen type Ⅰ and Ⅲ procollagen mRNA in primary cultured rat hepatocytes before and after stimulated by platelet-derived growth factor (PDGF) Effect of verapamil, a calcium antagonist, on this effect. Results: The hepatocytes cultured for a certain period of time could synthesize collagen and express type Ⅰ and Ⅲ procollagen genes. PDGF can promote collagen synthesis and procollagen gene expression in hepatocytes. However, this stimulus is inhibited by verapamil. Conclusion: These studies provide experimental evidence for the involvement of hepatocytes in the formation of hepatic fibrosis.
