目的:从淋巴细胞凋亡和调控的角度探讨清肺渗湿汤治疗哮喘气道炎症的机制。方法:清洁级雄性Wistar大鼠60只,随机分为5组,每组12只,采用卵蛋白制作大鼠哮喘模型,用TUNEL法测定了各组大鼠肺组织中淋巴细胞凋亡的百分比,用免疫组化方法研究肺组织中淋巴细胞上凋亡抑制基因Bcl-2的蛋白表达。结果:清肺渗湿汤、地塞米松片、麻杏石甘加味汤均能提高哮喘大鼠肺组织中淋巴细胞的凋亡率,下调凋亡抑制基因Bcl-2的蛋白表达,这种作用以前两者显著。结论:促进炎症局部淋巴细胞的凋亡是清肺渗湿汤减轻哮喘气道炎症反应一个主要机制。
Objective: To explore the mechanism of Qingfei Shenshi Decoction in treating asthmatic airway inflammation from the perspective of lymphocyte apoptosis and regulation. METHODS: Sixty male Wistar rats of clean grade were randomly divided into 5 groups with 12 rats in each group. Rat asthma model was established using ovalbumin. The percentage of lymphocyte apoptosis in rat lung tissues was determined by TUNEL method. Immunohistochemistry was used to study the expression of apoptosis-suppressing gene Bcl-2 on lymphocytes in lung tissue. RESULTS: Qingfei Shenshi Decoction, Dexamethasone Tablets and Maxing Shigan Jiawei Decoction can increase the apoptosis rate of lymphocytes in asthmatic rats and down-regulate the expression of Bcl-2 protein. Both were significant before. Conclusion: Promoting the apoptosis of local lymphocytes in inflammation is one of the main mechanisms of Qingfei Shenshi Decoction in alleviating asthma airway inflammation.
