为了探讨抗癌基因p53、癌转移抑制基因nm23的表达产物在胃癌及其对应的淋巴转移灶和正常组织中的表达,应用抗鼠p53,nm23H1单克隆抗体,用亲和素生物素过氧化物酶复合物法测定了这两种蛋白在上述组织中的表达.结果,在胃癌原发灶中nm23蛋白阳性表达率为41%,p53蛋白阳性表达率为50%;在对应淋巴转移灶中nm23蛋白阳性表达率为11%,而p53蛋白阳性表达率为67%.非癌组织中nm23蛋白表达全部为阴性,p53蛋白阳性表达率为2/6.nm23蛋白在胃癌原发灶中的阳性表达率显著高于非癌组织中的阳性表达率,胃癌原发灶中的阳性表达率显著高于转移灶中的nm23蛋白阳性表达率,原发灶高分化胃癌中的nm23蛋白阳性表达率显著高于低分化胃癌中的nm23蛋白阳性表达率;p53蛋白在胃癌淋巴转移灶中的阳性表达率显著高于非癌组织中的阳性表达率.
In order to investigate the expression of the anti-oncogene p53 and the metastasis suppressor gene nm23 in gastric carcinoma and its corresponding lymph metastases and normal tissues, anti-mouse p53, nm23H1 monoclonal antibody, and avidinbiotin were used. Peroxidase complex method was used to determine the expression of these two proteins in the above tissues. Results: The positive expression rate of nm23 protein was 41% in gastric cancer primary lesions, and the positive rate of p53 protein was 50%. The positive rate of nm23 protein was 11% in corresponding lymph node metastases, and the positive rate of p53 protein was 67%. . The expression of nm23 protein was negative in non-cancer tissues, and the positive rate of p53 protein was 2/6. The positive expression rate of nm23 protein in primary gastric cancer was significantly higher than that in non-cancer tissue. The positive expression rate of nm23 protein in primary tumor of gastric cancer was significantly higher than that of nm23 protein in metastasis, and the primary tumor was high. The positive expression rate of nm23 protein in differentiated gastric cancer was significantly higher than that in poorly differentiated gastric cancer. The positive rate of p53 protein in lymph node metastasis of gastric cancer was significantly higher than that in non-cancer tissue.
