目的:考察丹参总酚酸在裸鼠皮肤中的代谢稳定性,为丹参酚酸类成分的透皮制剂研究提供科学依据。方法:HPLC法测定生理盐水对照组和裸鼠皮肤匀浆实验组在32℃,300 r/min条件下保温24 h丹参总酚酸中丹酚酸B的含量,评价丹参总酚酸在裸鼠皮肤中的代谢稳定性。结果:32℃,300 r/min条件下24 h内丹酚酸B在生理盐水对照组及裸鼠皮肤匀浆组降解百分率分别为9.98%、15.92%(P<0.05)。丹酚酸B在生理盐水中的降解过程符合一级动力学方程,半衰期为66.87 h。结论:丹参总酚酸在裸鼠皮肤中不稳定,具有较明显的代谢作用。
OBJECTIVE: To investigate the metabolic stability of salvianolic acid in the skin of nude mice and to provide a scientific basis for the study of percutaneous preparation of salvianolic acids. Methods: The content of salvianolic acid B in Salvia miltiorrhiza total phenolic acid was determined by HPLC method in the control group of normal saline and the skin homogenate of nude mice at 32 ℃ and 300 r / min for 24 h, Metabolic stability in the skin. Results: The degradation rates of endosulfanilic acid B within 30 min at 300 ℃ were highest at 9 ℃ and 15.92% respectively (P <0.05). The degradation of salvianolic acid B in physiological saline conformed to the first-order kinetic equation with a half-life of 66.87 h. Conclusion: Salvia total phenolic acid in the skin of nude mice is unstable, with obvious metabolic effects.
