目的采用HPLC法测定并比较分析藤合欢中山柰苷的量。方法采用甲醇超声处理方法提取藤合欢,HPLC法测定山柰苷的量,色谱柱为Agilent Zorbax Eclipse Plus C_(18)柱(250 mm×4.6 mm,5μm),甲醇-0.01 mol/L磷酸二氢钾溶液-冰醋酸水溶液(40:60:1.5)等度洗脱,体积流量1.0 mL/min,检测波长254 nm,柱温30℃。结果线性范围在0.092 7~0.926 7μg(r=0.999 8),平均加样回收率为100.8%,RSD为1.1%(n=6)。结论该方法简便、准确,重复性好,可用于藤合欢质量控制;不同产地的藤合欢样本质量存在差异,贮藏时间越久,其所含的黄酮类成分越少。
OBJECTIVE To determine and compare the content of nerolidol in Acacia cacti by HPLC. Methods The extract of Acacia cuspidatum was extracted by ultrasonic method with methanol. The amount of behenyl glycosides was determined by HPLC. The column was Agilent Zorbax Eclipse Plus C 18 column (250 mm × 4.6 mm, 5 μm), methanol-0.01 mol / L dihydrogen phosphate Potassium acetate solution (40:60: 1.5), the volume flow rate was 1.0 mL / min, the detection wavelength was 254 nm and the column temperature was 30 ℃. Results The linearity ranged from 0.092 7 to 0.926 7 μg (r = 0.999 8) with an average recovery of 100.8% and a RSD of 1.1% (n = 6). Conclusion The method is simple, accurate and reproducible. It can be used for the quality control of Acacia canense. There are differences in the quality of Acacia samples from different areas. The longer the storage time, the less flavonoids it contains.
