本文比较了双核菌丝制备的香菇原生质体在不同培养基上的再生率,观察了原生质体再生过程.以存活率37%的剂量进行紫外线诱变处理后,选出102份再生克隆,从中选出在CM培养基和木屑培养基上菌丝长速较快的材料23份,进行栽培试验,其中10份产量提高,出菇较早.在随后的二次或三次组织分离后代的栽培试验中,大部分材料保持了增产、早熟的性状,经显著性测验.其中2份材料的产量显著提高.本研究初步建立了用原生质体诱变技术选育香菇菌株的试验程序.
In this paper, we compared the regeneration rate of protoplasts of Lentinula edodes on different media, and observed the regeneration process of protoplasts.Selecting 102 regenerated clones after UV mutagenesis with the survival rate of 37% In CM medium and sawdust medium, 23 hyphae fast-growing materials were cultivated, of which 10 were increased and fruiting earlier, and in the subsequent cultivation test of the second or third tissue separation progenies , Most of the materials maintained the yield-increasing, precocious traits by the significant test, in which the yield of two materials increased significantly.In this study, the experimental procedure of breeding lentinus by protoplast mutagenesis was preliminarily established.
