[摘 要]目的 测定 Z37株的全基因组序列,了解我国肾综合征出血热疫苗株分子基础,分析其遗传学差异,为疫苗生产提供分子依据。方法 设计出针对各片段的特异性引物,用PCR方法从Z37病毒感染的细胞提取总RNA,逆转录扩增产物克隆T载体纯化后测序,应用DNASTAR软件比较分析。结果Z37株全基因组由L片段6530个,M3651个和S1754个核苷酸组成,分别编码2151、1133和429个氨基酸,与同型病毒间同源率高达95.2%-99.5%,绘出了3个片段的核苷酸和氨基酸的系统发生树。结论 为研究疫苗毒株的生物学特性和致病机理等提供了分子基础。
[Abstract] Objective To determine the whole genome sequence of Z37 strain and to understand the molecular basis of the vaccine against Hemorrhagic Fever with Renal Syndrome in China. The genetic differences were analyzed to provide a molecular basis for vaccine production. Methods Specific primers for each fragment were designed. Total RNA was extracted from Z37 virus infected cells by PCR. The reverse transcription amplification product was cloned and sequenced. The T vector was sequenced and analyzed by DNASTAR software. Results The whole genome of Z37 strain consisted of 6530 L fragments, M3651 and S1754 nucleotides encoding 2151, 1133 and 429 amino acids, respectively. The homology of Z37 was 95.2% -99.5% A phylogenetic tree of 3 fragments of nucleotides and amino acids. Conclusion The molecular basis for studying the biological characteristics and pathogenicity of vaccine strains is provided.
