本文报告人体恶性间皮瘤细胞系AMS—82的建株过程及其生物学特性标本来源于一62岁女性病人。经剖腹探查及病理活检诊断为恶性间皮瘤。经贴壁静置培养目前已稳定传代生长三年多。该细胞系在相差镜下观察多数为长棱形细胞,有丰富的伪足及微绒毛。H、E、染色巨瘤细胞及核分裂相多见。传代三天后呈对数生长,倍增时间43小时,分裂指数82%。染色体分析提示该细胞主要为4倍体及超4倍体细肿。组化分析提示该细胞系能产生透明质酸。用BALB/C无胸腺裸鼠作肿瘤移植试验,皮下注射5×10~6细胞后4天长出瘤块,移植瘤的病理特征、组化分析与原始接种的肿瘤相同。将该细胞系在液氮中长期冻存,复苏均获成功。
This article reports that the process of constructing the human malignant mesothelioma cell line AMS-82 and its biological characteristics were derived from a 62-year-old female patient. After laparotomy and biopsy, it was diagnosed as malignant mesothelioma. The adherent static culture has now grown steadily for more than three years. Most of the cell lines observed under phase-contrast microscope were long prismatic cells with abundant pseudopodia and microvilli. H, E, stained giant tumor cells and mitoses are more common. Logarithmic growth was observed after three days of passage, with a doubling time of 43 hours and a division index of 82%. Chromosomal analysis suggested that the cells were mainly tetraploid and hypertetraploid. Histochemical analysis suggested that this cell line produced hyaluronic acid. BALB/C athymic nude mice were used for the tumor transplantation test. Tumor masses were grown 4 days after subcutaneous injection of 5×10-6 cells. The pathological features and histochemical analysis of the transplanted tumors were the same as those of the original tumors. The cell line was cryopreserved in liquid nitrogen for a long period of time and the recovery was successful.
