目的研究人表皮生长因子受体2(HER-2)、Ki-67、基质金属蛋白酶-9(MMP-9)的表达与脑膜瘤肿瘤组织病理分级的关系,探讨其临床病理学意义。方法选取脑膜瘤手术切除标本33份,采用免疫组化法(IHC)检测石蜡包埋组织标本中HER-2、Ki-67、MMP-9蛋白的表达情况,采用原位杂交方法(ISH)检测脑膜瘤石蜡包埋组织标本中HER-2、MMP-9编码的m RNA(HER-2 m RNA、MMP-9 m RNA)的表达情况,并用图象分析法进行形态学定量。结果恶性组和良性组HER-2、Ki-67、MMP-9蛋白的表达差异均有统计学意义(P<0.05)。良性复发组和良性非复发组的MMP-9 m RNA、HER-2 m RNA的表达差异有统计学意义(P<0.05)。结论 HER-2、Ki-67、MMP-9在脑膜瘤的表达与病理分级呈正相关,很可能是良恶性脑膜瘤鉴别诊断中有辅助作用的指标之一。HER-2、MMP-9在脑膜瘤的表达可能与脑膜瘤复发有关。HER-2及MMP-9较高表达可能与复发的良性脑膜瘤恶变有关。
Objective To investigate the relationship between the expression of human epidermal growth factor receptor 2 (HER-2), Ki-67 and matrix metalloproteinase-9 (MMP-9) and the histopathological grade in meningiomas, and to explore its clinicopathological significance. Methods 33 cases of meningiomas were selected for surgical resection. The expressions of HER-2, Ki-67 and MMP-9 in paraffin-embedded tissues were detected by immunohistochemistry (IHC). The expression of HER-2, The expression of HER-2 and MMP-9-encoded m RNA (HER-2 m RNA, MMP-9 m RNA) in paraffin-embedded tissue samples of meningioma was measured and morphologically quantified by image analysis. Results The expressions of HER-2, Ki-67 and MMP-9 in malignant group and benign group were significantly different (P <0.05). The expression of MMP-9 mRNA and HER-2 m RNA in benign relapse group and benign non-relapse group were significantly different (P <0.05). Conclusions The expression of HER-2, Ki-67 and MMP-9 in meningioma is positively correlated with the pathological grade, which is probably one of the auxiliary factors in the differential diagnosis of benign and malignant meningioma. The expression of HER-2 and MMP-9 in meningioma may be related to the recurrence of meningioma. The higher expression of HER-2 and MMP-9 may be related to the recurrence of benign meningioma.
