目的 选择简单的方法筛选“渗漏”型SCID小鼠。 方法 采用 3种酶联免疫吸附法 (双抗夹心ELISA、双抗夹心间接ELISA及生物素 亲和素夹心ELISA)测定SCID小鼠血清中IgG含量。 结果 生物素 亲和素夹心ELISA法灵敏度最高 ,用此法共检测了 4 1只 6~ 8周龄小鼠 ,其中 4 0只的IgG含量为 0 2 4± 0 16mg L ;只有 1只小鼠IgG含量为 2 3mg L ,渗漏比例为 2 4 %。 结论 用生物素 亲和素夹心ELISA法对SCID小鼠“渗漏”进行初筛 ,是简便、敏感、可行的方法
Objective To select a simple method to screen “leaky” SCID mice. Methods Serum IgG levels in SCID mice were determined by three enzyme-linked immunosorbent assays (double-antibody sandwich ELISA, double-antibody sandwich ELISA and biotin-avidin sandwich ELISA). Results The biotin-avidin sandwich ELISA method had the highest sensitivity. Using this method, 4 1 mice aged 6-8 weeks were screened, of which 40 had IgG of 0 24 ± 0 16 mg L; only 1 mouse IgG content of 23mg L, leakage ratio of 24%. Conclusion Preliminary screening of SCID mouse “leakage” with biotin-avidin sandwich ELISA is a simple, sensitive and feasible method
