目的研究在不同的脱水时间下,大鼠脑组织视上核中Lipocalin-2基因的变化情况。方法 60日龄雄性SD大鼠40只,随机分为脱水1d、2d、3d组和正常饮水对照组。脱水期满后处死动物,取脑组织视上核部分,采用RT-PCR和Western-blot检测Lipocalin-2的表达。结果与对照组相比,脱水2d、3d组视上核组织中Lipocalin-2的mRNA和蛋白表达水平都显著升高(P﹤0.05)。结论随着脱水时间的延长,Lipocalin-2在视上核组织中的含量逐渐增加,具有明显的时间-效应关系。提示了在机体脱水等原因导致的体液代谢混乱情况下,Lipocalin-2可能具有重要的生物学功能。
Objective To study the changes of Lipocalin-2 gene in supraoptic nucleus of rat brain under different dehydration time. Methods Forty male SD rats aged 60 days were randomly divided into two groups: dehydrated for 1 day, 2 days and 3 days, and normal drinking water control group. Animals were sacrificed at the expiration of the dehydration, and the supraoptic nucleus of brain tissue was taken. The expression of Lipocalin-2 was detected by RT-PCR and Western-blot. Results Compared with the control group, the mRNA and protein expression of Lipocalin-2 in the supraoptic nucleus on day 2 and day 3 after dehydration were significantly increased (P <0.05). Conclusion With the prolongation of dehydration, the content of Lipocalin-2 in supraoptic nucleus gradually increases with a significant time-effect relationship. Suggesting that Lipocalin-2 may have important biological functions in the body fluid metabolism disorders caused by dehydration and other causes.
