目的 :研究酿酒酵母细胞表面展示的人衰变加速因子 (hDAF) ,是否具有天然衰变加速因子的补体抑制活性。方法 :先用正常人血清处理酵母细胞EBY10 0 ,然后用流式细胞仪检测酵母细胞表面C5b 9复合物的沉积和hDAF在酵母细胞表面的表达。结果 :酵母细胞EBY10 0可直接活化人血清中的补体成分 ,导致酵母细胞表面C5b 9的沉积 ;而表达DAF的酵母细胞表面C5b 9的沉积明显减少。结论 :酵母细胞表面展示的DAF可正确折叠 ,并表现出DAF分子的补体抑制活性
Objective: To investigate whether human decay accelerating factor (hDAF) displayed on Saccharomyces cerevisiae cell surface has the complement inhibiting activity of natural decay accelerating factor. Methods: Yeast cells EBY10 0 were treated with normal human serum, then the deposition of C5b 9 complex on yeast cells and the expression of hDAF on the surface of yeast cells were detected by flow cytometry. RESULTS: EBY10 yeast cells directly activated human serum complement components, leading to the deposition of C5b9 on yeast cells, while the deposition of C5b9 on yeast cells expressing DAF was significantly reduced. Conclusion: DAF displayed on the surface of yeast cells can be correctly folded and exhibit the complement inhibitory activity of DAF molecules
