以大豆为外源DNA供体,用浸种及幼苗浇灌法和花粉管通道法直接将大豆总DNA导人受体水稻,经常规栽培获得水稻后代种子。采用薄层等电聚焦电泳和十二烷基硫酸钠-聚丙烯酸胺凝胶电泳法,分析了经大豆总DNA处理得到的水稻后代种子中的清蛋白、球蛋白、醇溶蛋白和谷蛋白。结果显示,大豆DNA可能引起水稻种子贮藏蛋白的变异。变异现象主要表现在两个方面;一是总条带数不变,但部分条带的迁移年与对照相比出现差异;再则就是出现了球蛋白、醇溶蛋白和谷蛋白条带增加的现象。
Soybean as an exogenous DNA donor, with soaking and seedling irrigation method and pollen tube pathway directly to the total soybean DNA introduced to the recipient of rice, the conventional cultivation of rice progeny seeds. The albumin, globulin, gliadin and glutelin in seeds of rice progenies derived from soybean total DNA treatment were analyzed by thin layer isoelectric focusing electrophoresis and sodium dodecyl sulfate - polyacrylamide gel electrophoresis. The results showed that soybean DNA may cause the variation of storage protein in rice seeds. Variation phenomenon mainly in two aspects: First, the total number of strips unchanged, but some of the strip migration year compared with the control there are differences; then there is the emergence of globulin, gliadin and gluten bands increased phenomenon.
