目的:探讨中药复方益糖康对链脲佐菌素(STZ)致糖尿病模型大鼠背根神经节(DRG)中NaV1.8蛋白及mRNA表达的影响。方法:将健康雄性Wistar大鼠40只,体重200-250g,随机分为空白对照组、模型组和益糖康组,其中,模型组和益糖康组采用STZ 55mg/kg腹腔注射复制糖尿病大鼠模型,然后分别给予安慰剂和益糖康5mL.次-1.天-1灌胃,6周后,采集大鼠背根神经节(DRG)标本,用免疫组化和RT-PCR方法分别检测其NaV1.8蛋白和mRNA表达水平。结果:模型组和益糖康组大鼠DRG中NaV1.8蛋白表达灰度值分别为102.19±10.62,134.04±6.56,mRNA表达积分吸光度比值分别为0.219±0.031,0.137±0.014。模型组大鼠DRG中NaV1.8蛋白和mRNA表达水平均显著上调,益糖康组较模型组NaV1.8蛋白和mRNA表达水平均显著降低。结论:中药复方益糖康可能对NaV1.8通道有阻断作用,是其改善糖尿病痛性神经病症状的机制之一。
Objective: To investigate the effect of Yikangkang, a traditional Chinese medicine, on the expression of NaV1.8 protein and mRNA in DRG of streptozotocin (STZ) -induced diabetic rats. Methods: Forty healthy male Wistar rats weighing 200-250 g were randomly divided into blank control group, model group and Yixikang group. The model group and the Yixikang group were treated with STZ 55 mg / kg intraperitoneally The rats were then given placebo and Yikang Kang 5mL, times -1, day -1, respectively. After 6 weeks, the dorsal root ganglion (DRG) samples of rats were collected and the expression of DRG was detected by immunohistochemistry and RT-PCR respectively The level of NaV1.8 protein and mRNA expression was detected. Results: The gray value of NaV1.8 protein expression in the DRG of the model group and the Yixikang group were 102.19 ± 10.62 and 134.04 ± 6.56, respectively, and the ratios of mRNA expression integral absorbance were 0.219 ± 0.031,0.137 ± 0.014, respectively. The expression level of NaV1.8 protein and mRNA in DRG of model group were significantly increased, while the expression level of NaV1.8 protein and mRNA in the model group was significantly lower than that of model group. Conclusion: Yikangkang, a traditional Chinese medicine compound, may block the channel of NaV1.8, which is one of the mechanisms to improve the symptoms of diabetic pain neuropathy.
