目的探讨类固醇激素对宫颈癌HeLa、肝癌细胞HEPG-2细胞端粒酶活性影响。方法采用MTT法检测类固醇激素作用前后HeLa、HEPG-2细胞凋亡情况;采用PCR-ELISA方法检测类固醇激素作用前后HeLa、HEPG-2细胞端粒酶活性的改变。结果乙烯雌酚不同程度抑制HeLa、HEPG-2细胞凋亡,增强HeLa、HEPG-2细胞端粒酶活性;而丙酸睾丸酮对此影响结果不显著。结论乙烯雌酚能不同程度激活端粒酶的活性,从而抑制肿瘤细胞的凋亡。
Objective To investigate the effect of steroid hormones on telomerase activity in HeLa and HEPG-2 cells. Methods MTT assay was used to detect the apoptosis of HeLa and HEPG-2 cells before and after steroid treatment. The changes of telomerase activity in HeLa and HEPG-2 cells before and after steroid treatment were detected by PCR-ELISA. Results Diethylstilbestrol inhibited the apoptosis of HeLa and HEPG-2 cells to varying degrees, and enhanced the telomerase activity of HeLa and HEPG-2 cells. However, the effect of testosterone propionate on HeLa and HEPG-2 cells was not significant. Conclusion Diethylstilbestrol can activate the activity of telomerase to some degree and inhibit the apoptosis of tumor cells.
