目的:研究氯化铝对体外培养大鼠睾丸支持细胞的雄激素结合蛋白(ABP)mRNA表达的影响。方法:体外培养大鼠睾丸支持细胞为模型,分为对照组、低剂量组、中剂量组、高剂量组(0,300,400,500μmol/L)氯化铝染毒睾丸支持细胞,培养24h后采取MTT法检测细胞活性,RT-PCR法检测ABP mRNA的表达水平。结果:随着氯化铝染毒剂量的增大,ABP mRNA的表达下降。高剂量染毒组与对照组相比较,ABP mRNA的表达差异具有统计学意义(P<0.05)。结论:氯化铝对体外培养的睾丸支持细胞具有毒性作用,抑制ABP mRNA的表达,从而损伤支持细胞功能,导致生殖功能障碍。
AIM: To investigate the effect of aluminum chloride on the expression of androgen-binding protein (ABP) mRNA in the cultured rat sertoli cells. Methods: Sertoli cells were cultured in vitro. The cells were divided into control group, low dose group, medium dose group and high dose group (0, 300, 400 and 500μmol / L) aluminum chloride for testis supportive cells. MTT assay The activity of ABP mRNA was detected by RT-PCR. Results: The expression of ABP mRNA decreased with the increase of the dose of aluminum chloride. Compared with the control group, the expression of ABP mRNA in high-dose exposure group was statistically significant (P <0.05). CONCLUSION: Aluminum chloride has toxic effect on cultured testicular sertoli cells and inhibits the expression of ABP mRNA, thus impairing the function of supporting cells and leading to reproductive dysfunction.
