目的研究lncRNA MUC5B-AS1基因在肺腺癌中的表达情况及对肺腺癌细胞系生物学行为的影响。方法利用RT-PCR、qRT-PCR检测肺腺癌细胞系中MUC5B-AS1基因的表达情况;构建MUC5B-AS1过表达载体,通过转染构建MUC5B-AS1过表达细胞系;采用CCK-8检测肺腺癌细胞的增殖情况;应用Transwell小室检测MUC5B-AS1过表达对肺腺癌细胞系迁移和侵袭的影响。结果检测了MUC5B-AS1在4株肺腺癌细胞系(A549、SPCA1、H1975和H1299)和1株肺正常上皮细胞系(HBE)中的表达情况,发现MUC5B-AS1在H1299细胞系中表达最低,同时A549细胞系表达最高;通过转染构建MUC5B-AS1过表达H1299、A549细胞系,利用qRT-PCR检测细胞系中MUC5B-AS1基因的表达,与对照组比较,MUC5B-AS1基因在H1299、A549细胞系均升高,且有统计学差异(P<0.05);CCK-8检测细胞增殖,与对照组比较过表达MUC5B-AS1对H1299、A549细胞生长并无影响(P>0.05);应用Transwell小室检测MUC5B-AS1过表达对肺腺癌细胞系迁移和侵袭,过表达MUC5B-AS1促进H1299、A549细胞的迁移和侵袭,差异有统计学意义(P<0.05)。结论 MUC5B-AS1可能在肺癌的迁移和侵袭发挥重要作用,但对肺腺癌细胞的增殖能力没有显著影响。
Objective To study the expression of lncRNA MUC5B-AS1 in lung adenocarcinoma and its effect on the biological behavior of lung adenocarcinoma cell lines. Methods The expression of MUC5B-AS1 gene in lung adenocarcinoma cell lines was detected by RT-PCR and qRT-PCR. MUC5B-AS1 overexpression vector was constructed and transfected into MUC5B-AS1 overexpressing cell line. CCK-8 was used to detect lung Proliferation of adenocarcinoma cells. The effect of MUC5B-AS1 overexpression on the migration and invasion of lung adenocarcinoma cell lines was detected by Transwell chamber. Results The expression of MUC5B-AS1 in four lung adenocarcinoma cell lines (A549, SPCA1, H1975 and H1299) and one lung normal epithelial cell line (HBE) was detected and MUC5B-AS1 was found to be the lowest in H1299 cell line , And A549 cell line was the highest. The expression of MUC5B-AS1 gene was detected by qRT-PCR in H1299 and A549 cell lines transfected with MUC5B-AS1. Compared with the control group, MUC5B- (P <0.05). The proliferation of A549 cells was detected by CCK-8. Compared with control group, overexpression of MUC5B-AS1 had no effect on the growth of H1299 and A549 cells (P> 0.05) The transwell chamber was used to detect MUC5B-AS1 overexpression in lung adenocarcinoma cell line Migration and invasion, MUC5B-AS1 overexpression promoted H1299, A549 cell migration and invasion, the difference was statistically significant (P <0.05). Conclusion MUC5B-AS1 may play an important role in the migration and invasion of lung cancer, but has no significant effect on the proliferation of lung adenocarcinoma cells.
