目的:建立测定栀子金花丸中盐酸小檗碱和绿原酸含量的毛细管电泳法。方法:采用未涂层弹性融硅石英毛细管柱(60 cm×75μm ID,有效长度52 cm);分别以0.2 mol.L-1磷酸二氢钠溶液+无水乙醇(50∶50)(pH 5.50)、50 mmol.L-1硼砂溶液为运行缓冲液;分离电压依次为21 kV,12 kV;重力进样10 s(高度15 cm);检测波长依次为265 nm,327 nm。结果:盐酸小檗碱和绿原酸浓度分别在11.0~44.0 mg.L-1,3.6~14.4 mg.L-1具有良好的线性关系(r均为0.999 3),平均加样回收率为100.0%,98.1%,方法精密度RSD 1.72%,0.96%(n=6)。栀子金花丸中盐酸小檗碱的平均含量3.39 mg.g-1;绿原酸的平均含量为0.88 mg.g-1。结论:该方法准确、灵敏、快速,可用于栀子金花丸中盐酸小檗碱和绿原酸的质量控制研究。
Objective: To establish a capillary electrophoresis method for the determination of berberine hydrochloride and chlorogenic acid in Zhizi Jinhua Wan. Methods: Uncoated flexible fused silica capillary column (60 cm × 75 μm ID, effective length of 52 cm) were prepared by using 0.2 mol·L-1 sodium phosphate monobasic solution and absolute ethanol (50:50) ) And 50 mmol·L-1 borax solution were used as running buffer. The separation voltages were 21 kV and 12 kV, respectively. The gravimetric injection time was 10 s (height 15 cm). The detection wavelength was 265 nm and 327 nm, respectively. Results: Berberine hydrochloride and chlorogenic acid had good linearity (r = 0.999 3) in the range of 11.0-44.0 mg.L-1, 3.6-6.4 mg.L-1, respectively. The average recoveries of berberine and chlorogenic acid were 100.0 %, 98.1%, the method precision RSD 1.72%, 0.96% (n = 6). The average content of berberine hydrochloride in Gardenia Jinhua Wan was 3.39 mg.g-1; the average content of chlorogenic acid was 0.88 mg.g-1. Conclusion: The method is accurate, sensitive and rapid and can be used for quality control of berberine hydrochloride and chlorogenic acid in Zhizi Jinhua Wan.
