目的:观察β-连环蛋白表达的变化趋势,从而了解β-连环蛋白在烟雾吸入性肺损伤大鼠中的活化情况及意义。方法48只Wistar大鼠均分为两组,A组建立烟雾吸入性肺损伤模型,B组为正常对照组。建模后第3、7、14天从两组随机各选取8只处死,取出左肺,用显微镜进行病理检查,用RT-PCR法检测β-连环蛋白表达水平。结果病理检查确定烟雾吸入性肺损伤模型制备成功。β-连环蛋白的 mRNA表达水平在致伤后第3天高于正常组,于第7天表达水平达到最高(均P0.05)。结论烟雾吸入性肺损伤后以β-连环蛋白为关键信号分子的Wnt/β-连环蛋白信号通路激活,且该信号分子在受损组织中可能发挥增殖、修复的作用。“,”Objective To investigate the expression of Wnt/β-catenin in smoke inhalation lung injury and signioficance thereof. Methods Forty-eight Wistar rats were randomly diveded into 2 equal groups: Group A undergoing smoke inhalation lung injury so as to establish smoke inhalation lung injury model, and Group B used as normal controls. On days 3, 7, and 14 after the establishment of model, 8 rats randomly selected from each group were killed with their lungs taken out to undergo microscopic examination to observe the pathological changes and undergo RT-PCR to test the mRNA expression of β-catenin. Results Pathological examination shoed that the smoke inhalation lung injury model was successfully made. The mRNA expression level ofβ-catenin in was significantly higher in Grouip A than in Group B on days 3 and 7 after the model establishment (both P 0.05). Conclusion Wnt/β-catenin signaling pathway, with β-catenin as the key signal molecule is activated in the smoke inhalation lung injury, and it plays a role in the proliferation and repair of the damaged lung tissue.
