目的:研究参附注射液对大鼠心肌肥大模型micro RNA-199a-5p的调控机制。方法:60只健康SD大鼠分成3组,模型组和参附注射液组行腹主动脉缩窄术建立心肌肥大模型,假手术组腹主动脉不作缩窄。参附注射液组腹腔注射参附注射液6.0m L·kg~(-1)·d~(-1);模型组和假手术组均腹腔注射0.9%氯化钠溶液6.0m L·kg~(-1)·d~(-1),连续12周。采用Realtime RT-PCR方法检测大鼠心肌micro RNA-199a-5p的表达。利用生物信息学方法分析micro RNA-199a-5p的候选靶基因,并用Western blot方法检测靶基因HSP70的蛋白表达水平。结果:与假手术组比较,模型组大鼠心肌micro RNA-199a-5p上调(P<0.05);参附注射液组大鼠心肌micro RNA-199a-5p表达量较模型组下调(P<0.05);参附注射液组HSP70蛋白表达较假手术组和模型组显著增加(P<0.01)。结论:micro RNA-199a-5p在心肌肥大中可能发挥重要作用,参附注射液可能通过抑制micro RNA-199a-5p的上调,增加HSP70的蛋白表达水平,抑制心肌肥大大鼠心室重构,从而改善大鼠心功能。
Objective: To study the regulatory mechanism of Shenfu Injection on rat cardiac hypertrophy model micro RNA-199a-5p. Methods: Sixty healthy SD rats were divided into three groups. The model group and the Shenfu injection group were given abdominal aortic constriction to establish the model of cardiac hypertrophy. The abdominal aorta of the sham operation group was not narrowed. Shenfu injection group was given intraperitoneal injection of Shenfu Injection 6.0m L · kg -1 · d -1; 0.9% sodium chloride solution 6.0ml · kg -1 was injected intraperitoneally into model group and sham group (-1) · d ~ (-1) for 12 weeks. Realtime RT-PCR was used to detect the expression of micro RNA-199a-5p in rat myocardium. Candidate target genes of microRNA-199a-5p were analyzed by bioinformatics methods, and the protein expression level of HSP70 was detected by Western blot. Results: Compared with the sham operation group, the micro RNA-199a-5p in the model group was significantly increased (P <0.05); the expression of micro RNA-199a-5p in the myocardium of Shenfu injection group was significantly lower than that in the model group ); HSP70 protein expression in Shenfu injection group was significantly higher than that in sham operation group and model group (P <0.01). Conclusion: microRNA-199a-5p may play an important role in myocardial hypertrophy. Shenfu injection may inhibit the upregulation of micro RNA-199a-5p, increase the protein expression of HSP70, and inhibit ventricular remodeling in rats with cardiac hypertrophy. Improve heart function in rats.
