目的 研究胰腺癌组织中smad4 mRNA ,TGF β1 和TGF βR1 的表达及其生物学意义。方法 采用原位杂交方法检测癌及癌旁组织的smad4 mRNA ,免疫组化方法检测TGF β1 和TGF βR1 。结果 5 3例胰腺癌smad4 mRNA ,TGF β1 和TGF βR1 表达阳性率明显地低于 2 5例癌旁上皮 (均P <0 .0 5 )。高分化腺癌smad4 mRNA ,TGF β1 和TGF βR1 表达阳性率明显高于低分化腺癌 (P <0 .0 5~ 0 .0 1) ,无转移癌明显高于转移癌 (P <0 .0 5~ 0 .0 1)。三者在胰腺癌中表达阳性率均存在密切相关系。结论 smad4 mRNA ,TGF β1 和TGF βR1 表达与胰腺癌的发生发展、生物学行为和预后有密切关系 ,检测三者表达水平对早期发现胰腺癌及指导临床治疗和评估预后可能有较重要价值。
Objective To study the expression of smad4 mRNA, TGFβ1 and TGFβR1 in pancreatic carcinoma and its biological significance. Methods Smad4 mRNA was detected by in situ hybridization in situ hybridization and immunohistochemistry to detect TGF β1 and TGF β R1. Results The positive rates of smad4 mRNA, TGFβ1 and TGFβR1 in 53 cases of pancreatic cancer were significantly lower than those of 25 cases of paracancerous epithelium (all P <0.05). The positive rates of smad4 mRNA, TGFβ1 and TGFβR1 in well-differentiated adenocarcinoma were significantly higher than those in poorly differentiated adenocarcinoma (P <0.05-0.01) 5 ~ 0 .0 1). The three positive rates in pancreatic cancer are closely related. Conclusion The expression of smad4 mRNA, TGFβ1 and TGFβR1 are closely related to the development, biological behavior and prognosis of pancreatic cancer. Detecting the expression of these three genes may have important value in the early detection of pancreatic cancer, guiding the clinical treatment and evaluating the prognosis.
