观察多脏器功能不全综合征(MultipleOrganDysfunctionSyndrome,MODS)模型大鼠肝细胞线粒体功能及下法药物对其影响.方法:以改良NBT比色法代替2,6—二氯酚靛酚法测定急性感染性腹膜炎及肠系膜动脉缺血再灌流所致MODS模型大鼠肝细胞线粒体功能,并对LPS体外刺激大鼠肝细胞所致其线粒体功能改变及寒下药物对其影响进行观察.结果;体外实验结果表明,LPS刺激的大鼠肝细胞线粒体功能增强(P<0.05),大承气汤、承气合剂及大黄对LPS活化的大鼠肝细胞线粒体功能具抑制作用(P<0.05),MODS大鼠肝细胞线粒体功能较对照组明显升高(P<0.05),大承气汤灌胃治疗后其肝细胞线粒体功能较模型组下降(P<0.05)。结论:MODS发病或经体外LPS刺激活化后大鼠肝细胞线粒体功能均增强,以大承气汤为代表的寒下药物能抑制此线粒体功能增强作用,抑制线粒体的呼吸爆发,因此抑制超氧化阴离子的生成,达到保护组织脏器的作用。
To observe the mitochondrial function of hepatocytes in multiple organ dysfunction syndrome (MODS) model rats and the effects of the drugs under the law. Methods: The mitochondrial function of liver cells in MODS rat models induced by acute infectious peritonitis and mesenteric artery ischemia-reperfusion were determined by modified NBT colorimetric method instead of 2,6-dichlorophenol-indophenol method. The effects of LPS stimulation on rat liver The change of mitochondrial function caused by cells and the influence of cold medicine on it were observed. Results: In vitro, mitochondrial function of LPS-stimulated rat hepatocytes was enhanced (P <0.05). Dachengqi decoction, Chengqi mixture and rhubarb inhibited the mitochondrial function of LPS-activated rat hepatocytes (P (P <0.05). The function of mitochondria of hepatocytes in MODS rats was significantly higher than that in the control group (P <0.05). The mitochondrial function of hepatocytes in Dachengqi Decoction treatment group was lower than that in the model group ). CONCLUSION: The mitochondrial function of rat hepatocytes is enhanced after MODS onset or activation by LPS stimulation in vitro. Cold medicine, represented by Dachengqi Decoction, can inhibit the mitochondrial function enhancement and inhibit the respiratory burst of mitochondria, thus inhibiting the superoxide anion The formation of organs to protect the organization’s role.
