研究了HPLC测定蜂胶葡萄酒中8种有机酸的快速检测方法。优化得到色谱条件:色谱柱,Agilent Eclipse XDB-C18(5μm,4.6 mm×250 mm);流动相为15 mmol/L KH2PO4(用磷酸调p H至2.1)-甲醇(98∶2),流速0.8 m L/min;紫外检测波长205 nm和243 nm,柱温30℃,进样量10μL,等度洗脱。结果表明,8种有机酸的线性关系良好,相关系数在0.999 1~1之间;精密度、稳定性的RSD都低于3.08%;平均回收率93.60%~109.00%,定量限为0.001 2~0.015 mg/m L之间,各组分相对标准偏差(n=6)为0.23%~3.08%。该方法简便,具有较高的准确度和精密度,可应用于蜂胶葡萄酒中有机酸的检测。
The rapid determination of 8 organic acids in propolis wine by HPLC was studied. The mobile phase consisted of 15 mmol / L KH2PO4 (adjusted to pH 2.1 with phosphoric acid) -methanol (98: 2), and the flow rate was 0.8. The chromatographic column was Agilent Eclipse XDB-C18 m L / min; UV detection wavelength 205 nm and 243 nm, column temperature 30 ℃, injection volume 10μL, isocratic elution. The results showed that the linear relationships of the eight organic acids were good with the correlation coefficients of 0.999 1 ~ 1. The RSDs of precision and stability were all lower than 3.08%. The average recoveries were 93.60% ~ 109.00% and the limits of quantification were 0.001 2 ~ 0.015 mg / m L, the relative standard deviations (n = 6) of each component ranged from 0.23% to 3.08%. The method is simple, has high accuracy and precision, and can be applied to the detection of organic acids in propolis wine.
