目的:为了进一步研究蝉拟青霉总多糖和腺苷单用或分别联合顺铂(DDP)或阿霉素(ADM)在体外的抗肿瘤作用。方法:用MTT法检测DDP、ADM、总多糖、总腺苷单独及联合用药对人肺腺癌(PAA-1)细胞的生长抑制能力;用流式细胞仪检测其对体外培养细胞系细胞周期的影响;用光镜检查其对体外培养PAA-1细胞系集落形成率的影响;用流式细胞仪检测其对体外培养淋巴细胞亚群分化的影响。结果:DDP、ADM、总多糖、腺苷对人肺腺癌(PAA-1)细胞生长抑制的IC50分别为9.05μg/mL、36.21μg/mL、10.86mg/mL、7.81mg/mL。DDP或ADM和多糖联用对PAA-1细胞的生长抑制主要表现出拮抗效应;0.5mg/mL、0.25mg/mL腺苷与4μg/mL阿霉素联合作用时,显示出拮抗效应,而当8mg/mL腺苷与4μg/mL阿霉素联用时,显示协同效应;不同浓度腺苷单用与和DDP联合使用相比,对细胞生长影响无显著性差异;腺苷和ADM联合作用,随着腺苷浓度的增加,G0/G1期细胞逐渐减少,而G2/M期相对增加。对体外培养PAA-1细胞系集落形成率多糖组和阴性对照组未见明显差异,不同浓度腺苷组的集落形成率略低于阴性对照组,随着腺苷浓度逐渐增加,集落形成率逐渐下降。多糖对体外培养淋巴细胞亚群的分化无影响。结论:总多糖、腺苷具有一定的体外抗肿瘤作用,总多糖与DDP或ADM联用不能增加其体外的抗肿瘤作用,腺苷与ADM联用可增加其体外的抗肿瘤作用。
OBJECTIVE: To further study the antitumor effect of total polysaccharides and adenosine monophylla in vitro combined with cisplatin (DDP) or doxorubicin (ADM). Methods: The inhibitory effects of DDP, ADM, total polysaccharides and total adenosine alone and in combination on the growth of human lung adenocarcinoma (PAA-1) cells were detected by MTT assay. The cell cycle of the cultured cells was detected by flow cytometry The effect of light microscope on the colony formation rate of PAA-1 cells in vitro was examined by light microscopy. The effects on the differentiation of cultured lymphocyte subsets in vitro were detected by flow cytometry. Results: The IC50 of DDP, ADM, total polysaccharide and adenosine on human lung adenocarcinoma (PAA-1) cell growth were 9.05μg / mL, 36.21μg / mL, 10.86mg / mL and 7.81mg / mL respectively. DDP or ADM combined with polysaccharides mainly inhibited the growth of PAA-1 cells. Antagonism was observed when 0.5mg / mL and 0.25mg / mL adenosine combined with 4μg / mL doxorubicin, Adenosine 8mg / mL combined with 4μg / mL doxorubicin showed synergistic effect; different concentrations of adenosine alone and compared with the combination of DDP had no significant difference on cell growth; adenosine and ADM combined effect, with With the increase of adenosine concentration, G0 / G1 phase cells gradually decreased, while G2 / M phase increased. Colony formation rate of PAA-1 cells cultured in vitro showed no significant difference between the group and the negative control group, different concentrations of adenosine group colony formation rate slightly lower than the negative control group, with the adenosine concentration gradually increased, the rate of colony formation gradually decline. Polysaccharide had no effect on the differentiation of cultured lymphocyte subsets in vitro. CONCLUSION: Total polysaccharides and adenosine have anti-tumor effect in vitro. Total polysaccharide combined with DDP or ADM can not increase the anti-tumor effect in vitro. Combined use of adenosine and ADM can increase its anti-tumor effect in vitro.
