[目的]观察便塞通合剂对实验性便秘大鼠PGP9.5、SS的影响及其效果差异。[方法]将80只SD大鼠随机分为空白对照组、模型组、麻仁丸组、便塞通合剂组,每组20只,采用改良复方地芬诺酯法制作大鼠实验性便秘模型,除空白对照组和模型组不进行药物治疗外,麻仁丸组和便塞通合剂组分别给予麻仁丸0.3g·kg-1·d-1灌胃,便塞通合剂7.5g·kg-1·d-1灌胃。14d后处死全部大鼠,观察大鼠结肠组织病理学改变,采用免疫组织化学法检测结肠组织中PGP9.5、SS的表达水平。[结果]HE染色空白对照组大鼠结肠黏膜正常,模型组大鼠结肠黏膜有炎性细胞浸润,肌间神经丛神经细胞减少,可见空泡变性。通塞便合剂治疗后,近端结肠黏膜未见炎性细胞浸润,肌间神经纤维未见空泡变性。与对照组比较,模型组SS表达显著增加(P<0.05),PGP9.5表达显著减少(P<0.05);麻仁丸组和便塞通组均表现为下调SS的高表达状态,明显低于模型组(P<0.05),上调PGP9.5,明显高于模型组(P<0.05);而便塞通合剂组对其表达影响明显优于麻仁丸组(P<0.05)。[结论]便塞通合剂对便秘大鼠有较好的疗效,主要表现在控制黏膜炎症反应、降低组织中SS的表达水平,上调PGP9.5表达。
[Objective] To observe the effect of Huatong Tongji on PGP9.5 and SS in experimental constipation rats and its effect difference. [Method] Eighty SD rats were randomly divided into blank control group, model group, Maren pill group and Huishengtong agent group, with 20 rats in each group. Experimental constipation model was made by modified diphenoxylate method , Except for the blank control group and the model group without drug treatment, Maren pill group and the plug combined group were given Maren pill 0.3g · kg-1 · d-1 gavage, the plug and the synergist 7.5g · kg -1 · d-1 gavage. After 14 days, all the rats were sacrificed and the histopathological changes of the colon were observed. The expression of PGP9.5 and SS in colonic tissue was detected by immunohistochemistry. [Result] The colonic mucosa of rats in normal control group were normal, the inflammatory cells infiltrated in colonic mucosa of model group, and the number of neurons in myenteric plexus decreased. Tongsheyin mixture treatment, no inflammatory cell infiltration of the proximal colon mucosa, myenteric nerve fibers no vacuolar degeneration. Compared with the control group, the expression of SS in model group was significantly increased (P <0.05) and the expression of PGP9.5 was significantly decreased (P <0.05); Maren pill group and Compared with model group (P <0.05), PGP9.5 increased significantly (P <0.05), while that of Huayan Mixture group was significantly higher than that of Maren pill group (P <0.05). [Conclusion] Huitong Tonghe agent has good curative effect on constipation rats, mainly in controlling mucosal inflammation, reducing the expression of SS in the tissue and up-regulating the expression of PGP9.5.
