双孢蘑菇菌株的菌丝体酯酶同工酶检测法

用聚丙烯酰胺凝胶平板电泳法测定双孢蘑菇菌丝体的酯酶同工酶谱,表明同一菌株用马铃薯、葡萄糖、麦芽汁液体培养基(PDM)静止培养(24±1℃)45天的菌丝体酯酶同工酶与用该培养基摇床培养(23℃,71转/分)20天的菌丝体酯酶同工酶相同;酯酶提取液存放(4℃)的时间对酯酶同工酶谱的表现有很大的影响,即酯酶同工酶谱随酯酶提取液存放时间的延长而逐渐变糊乃至消失;双孢蘑菇种内菌株间的酯酶同工酶差异显著。与测定双孢蘑菇子实体组织的酯酶同工酶的方法相比较,测定双孢蘑菇菌丝体酯酶同工酶的方法具有简便、可靠、灵敏等优点,因而它为双孢蘑菇菌株的早期鉴定提供了一种有用的手段。 The esterase isozyme pattern of Agaricus bisporus mycelia was determined by polyacrylamide gel electrophoresis, indicating that the same strain was still cultured (24 ± 1 ° C) for 45 days in potato, glucose, and malt broth (PDM) Mycelia esterase isoenzymes were the same as mycelial esterase isoenzymes cultured in this medium for 20 days at 23 ℃ (71 rpm); the time of esterase extract storage (4 ℃) Esterase isozyme profile of the performance has a significant impact, that is, esterase isozyme profile with esterase extract storage time gradually become fuzzy or even disappear; Agaricus bisporus isolates Esterase between workers Enzymes significantly different.
