目的:采用超声法提取阿尔泰狗娃花中的多糖,并测定其含量。方法:采用紫外-可见分光光度法,以葡萄糖为对照品,苯酚-浓硫酸显色法测定阿尔泰狗娃花中多糖的含量。结果:总多糖在2.535-15.21μg/mL范围内呈良好线性关系,回归方程Y=0.0501X-0.0015(r=0.9998,n=6),阿尔泰狗娃花多糖含量为5.645%,RSD为0.65%。经方法学考察显示,稳定性、精密度、重现性均较好,平均回收率为96.86%,RSD为1.45%。结论:紫外分光光度法测定阿尔泰狗娃花中的多糖,操作简便、灵敏度高、重现性好,稳定性高,可作为阿尔泰狗娃花中多糖含量测定方法。
OBJECTIVE: To extract the polysaccharides from Altai dog pupa by ultrasonic method and determine its content. Methods: The content of polysaccharide in Altay dog flower was determined by UV - visible spectrophotometry with glucose as reference substance and phenol - concentrated sulfuric acid colorimetry. Results: The total polysaccharide showed a good linear relationship in the range of 2.535-15.21μg / mL. The regression equation was Y = 0.0501X-0.0015 (r = 0.9998, n = 6) . The methodological study showed that the stability, precision and reproducibility were good, the average recovery was 96.86%, RSD was 1.45%. Conclusion: UV spectrophotometry polysaccharide polysaccharide in the flower of Altai dog, easy to operate, high sensitivity, good reproducibility, high stability, can be used as the method of determination of polysaccharide content of Altai dog.
