目的根据前期工作中运用血浆蛋白质组学研究筛选出的一系列血浆候选差异蛋白指标,探索血浆中ATⅢ、VIL1和CLU表达含量在维吾尔族妇女宫颈病变进程中动态变化。方法收集82例新疆地区维吾尔族宫颈病变血浆样本(宫颈炎22例,CINⅡ~Ⅲ20例,宫颈癌早期20例,宫颈癌晚期20例),用酶联免疫法检测血浆中ATⅢ、VIL1和CLU的表达含量,分析其与宫颈病变进程的关系及相关性。结果 ATⅢ的表达含量随着宫颈病变进程逐渐下降,CLU和VIL1的表达含量随着宫颈病变的进程逐渐升高(P<0.05)。以宫颈炎组作为对照组,ATⅢ、VIL1和CLU的表达含量与CINⅡ~Ⅲ组、宫颈癌早期组比较差异均有统计学意义(P<0.05)。ATⅢ、VIL1和CLU表达含量之间的动态变化关联性分析差异无统计学意义。结论维吾尔族妇女宫颈癌发生发展与ATⅢ、VIL1和CLU的表达含量动态变化密切相关,可成为宫颈癌早期诊断的血浆候选预警蛋白标志物。
Objective To investigate the dynamic changes of plasma levels of ATⅢ, VIL1 and CLU in Uygur women with cervical lesions based on a series of plasma protein candidate studies using plasma proteomics. Methods Totally 82 Uygur cervical lesions (22 cases of cervicitis, 20 cases of CIN Ⅱ ~ Ⅲ, 20 cases of early cervical cancer and 20 cases of advanced cervical cancer) were collected from Xinjiang Uygur patients. The levels of AT Ⅲ, VIL1 and CLU in plasma were detected by enzyme-linked immunosorbent assay Expression, analysis of its relationship with the process of cervical lesions and correlation. Results The expression of AT Ⅲ gradually decreased with the progression of cervical lesions. The expression of CLU and VIL1 gradually increased with the progress of cervical lesions (P <0.05). Cervicitis group as a control group, AT Ⅲ, VIL1 and CLU expression levels compared with CIN Ⅱ ~ Ⅲ group, cervical cancer early group were significantly different (P <0.05). There was no significant difference in the dynamic change between ATⅢ, VIL1 and CLU expression levels. Conclusions The occurrence and development of cervical cancer in Uighur women are closely related to the dynamic changes of the expression of AT Ⅲ, VIL1 and CLU, which may be used as the early warning plasma protein candidate for early diagnosis of cervical cancer.
