目的:建立川贝母细胞团块悬浮培养体系,筛选出快速获取细胞团块增殖的最佳培养条件和激素配比。方法:以MS培养基为基本培养基,比较了接种量、激素配比和生长调节物质等对川贝母细胞团块悬浮培养的影响,并比较了细胞团块在不同培养条件下的生长情况,测定了增殖细胞团块中总生物碱含量。结果与结论:用川贝母细胞团块进行悬浮培养,其生长速度明显快于固体培养;细胞团块中总生物碱含量均高于市售川贝母和野生川贝母鳞茎;川贝母细胞团块悬浮培养最适的接种量为30 g/L,最佳培养基为MS+6-BA 2.0 mg/L+NAA 0.2 mg/L。
OBJECTIVE: To establish a suspension culture system of Fritillaria cirrhosa fibroblasts and screen out the best culture conditions and hormonal ratios for rapidly obtaining the cell mass proliferation. Methods: MS medium was used as the basic medium. The effects of inoculum size, hormone ratio and growth-regulating substances on the suspension culture of Fritillaria cirrhosa cells were compared. The growth of the cells in different culture conditions was compared , The total alkaloid content in the proliferating cell mass was measured. RESULTS AND CONCLUSION: The growth of Fritillaria cirrhosa cells in suspension was significantly faster than that in solid culture. The content of total alkaloids in the cell mass was higher than that of the commercially available Fritillaria cirrhosa and wild Fritillaria cirrhosa. The optimal inoculation amount for cell suspension culture was 30 g / L, and the best culture medium was MS + 6-BA 2.0 mg / L + NAA 0.2 mg / L.
