目的探讨胰腺癌血清Periostin(PN)水平与淋巴结转移的关系。方法采用ELISA检测血清中PN水平,并进行自制ELISA试剂盒的指标评价。结果 ELISA法检测PN的最小检出限为4.903 ng/ml,标准曲线方程式曲线拟合度R2=0.968。平均批内变异系数为7.6%,平均批间变异系数为13.15%,回收率为82%~92%。健康对照者31例的血清PN含量为(22.0±6.8)ng/ml,18例胰腺癌患者的血清PN含量为(37.9±20.9)ng/ml,18例有远处转移的胰腺癌患者的血清PN含量为(45.6±25.8)ng/ml。胰腺癌患者与健康对照者间有显著性差异(P<0.05),而18例胰腺癌患者与健康对照者和18例有远处转移的胰腺癌患者组间也有显著性差异(P<0.05)。结论 PN-ELISA试剂盒具有良好的灵敏性、特异性和准确性。PN与肿瘤转移有密切关系,且PN蛋白水平的检测可以用于判断胰腺癌和作为诊断胰腺癌是否出现淋巴结转移的指标。
Objective To investigate the relationship between serum Periostin (PN) level and lymph node metastasis in pancreatic cancer. Methods The levels of PN in serum were detected by ELISA and the indexes of homemade ELISA kit were evaluated. Results The minimum detectable limit of detection of PN by ELISA was 4.903 ng / ml. The curve fitting curve of standard curve was R2 = 0.968. The average intra-assay CV was 7.6%, the average inter-assay CV was 13.15% and the recovery was 82% -92%. The serum PN content of healthy controls was (22.0 ± 6.8) ng / ml in 31 cases, that of pancreatic cancer in 18 cases (37.9 ± 20.9) ng / ml, and that of 18 cases of distant metastasis The PN content was (45.6 ± 25.8) ng / ml. There were also significant differences between pancreatic cancer patients and healthy controls (P <0.05), but there were also significant differences between 18 pancreatic cancer patients and healthy controls and 18 patients with distant metastasis (P <0.05) . Conclusion PN-ELISA kit has good sensitivity, specificity and accuracy. PN and tumor metastasis are closely related, and the detection of PN protein levels can be used to determine the pancreatic cancer and pancreatic cancer as a diagnosis of lymph node metastasis or not.
