目的探讨糖氧剥夺(oxygen-glucose deprivation,OGD)对体外培养的大鼠星形胶质细胞窖蛋白1(caveolin-1,Cav-1)的影响。方法原代培养出生24h内SD大鼠大脑皮质星形胶质细胞,分为对照组和OGD组(给予星形胶质细胞6h的OGD和24h的再灌注损伤,建立OGD损伤模型)。用多重免疫荧光的方法检测星形胶质细胞Cav-1表达,实时荧光定量PCR和Western blot的方法检测星形胶质细胞Cav-1表达的变化。根据小干扰RNA(siRNA)的原理,将Cav-1siRNA转染星形胶质细胞,采用CCK-8的方法检测星形胶质细胞活力。结果大鼠大脑皮质星形胶质细胞Cav-1普遍表达。与对照组比较,OGD组星形胶质细胞Cav-1mRNA和蛋白表达量下降(P<0.05)。siRNA转染后,与对照组比较,OGD组星形胶质细胞活力下降(P<0.05)。结论 SD大鼠大脑皮质星形胶质细胞Cav-1表达的下调,导致OGD后细胞损伤加重,提示Cav-1对星形胶质细胞具有保护作用。
Objective To investigate the effect of oxygen-glucose deprivation (OGD) on caveolin-1 (Cav-1) in rat astrocytes. Methods Primary cultured rat cerebral cortical astrocytes 24 h after birth were divided into control group and OGD group (OGD given astrocytes for 6 hours and reperfusion injury for 24 hours and established OGD injury model). The expression of Cav-1 in astrocytes was detected by multiple immunofluorescence method. The expression of Cav-1 in astrocytes was detected by real-time fluorescence quantitative PCR and Western blot. According to the principle of small interfering RNA (siRNA), Cav-1 siRNA was transfected into astrocytes and the viability of astrocytes was detected by CCK-8. Results The expression of Cav-1 in cerebral cortex astrocytes in rats was widespread. Compared with the control group, the expression of Cav-1 mRNA and protein in astrocytes of OGD group decreased (P <0.05). After transfection, the viability of astrocytes in OGD group decreased compared with the control group (P <0.05). Conclusions The downregulation of Cav-1 expression in the astrocytes of the SD rats resulted in the increased cell injury after OGD, suggesting that Cav-1 may have a protective effect on astrocytes.
