目的:运用荟萃分析方法综合评价肿瘤坏死因子α基因启动子308位(TNF-α-308)G/A基因型与溃疡性结肠炎(UC)的相关性。方法:检索PubMed、EMBase、CNKI和万方数据库,获取TNF-α-308基因多态性与溃疡性结肠炎易感性的病例-对照研究,使用统一的表格提取数据,应用Review Manager4.2软件进行统计学处理。结果:检索文献中纳入14个病例-对照研究,其中欧洲8篇,美洲2篇,亚洲4篇(东亚3篇)。不按种族分层时,等位基因TNF2与UC易感性无相关性(OR1.32,95%CI0.84~2.07,P=0.22),按种族分层时,东亚人群等位基因TNF2与UC密切相关(OR3.12,95%CI1.44~6.77,P=0.004),欧洲人群等位基因TNF2与UC易感性无相关性(OR0.73,95%CI0.41~1.31,P=0.30),基因型(TNF2/1+TNF2/)vsTNF1/1者的UC易感性与等位基因TNF2者相似。结论:在东亚人群中TNF2等位基因是UC的危险因素,在欧洲人群中TNF-α-308的G/A基因多态性与UC无关。
OBJECTIVE: To evaluate the relationship between TNF-α-308 G / A genotype and ulcerative colitis (UC) using meta-analysis. Methods: PubMed, EMBase, CNKI and Wanfang database were searched for a case-control study of TNF-α-308 gene polymorphism and susceptibility to ulcerative colitis. Data were extracted using a uniform table and reviewed using Review Manager 4.2 software Statistical analysis. Results: A total of 14 case-control studies were included in the search of literature, including 8 in Europe, 2 in the Americas and 4 in Asia (3 in East Asia). When stratified by ethnicity, there was no correlation between allele TNF2 and susceptibility to UC (OR1.32,95% CI0.84-2.07, P = 0.22). When stratified by ethnicity, (OR3.12,95% CI 1.44 ~ 6.77, P = 0.004). No association was found between European population allele TNF2 and susceptibility to UC (OR 0.73, 95% CI 0.41 to 1.31, P 0.30) , Genotype (TNF2 / 1 + TNF2 /) vsTNF1 / 1 UC susceptibility and allele TNF2 were similar. CONCLUSIONS: The TNF2 allele is a risk factor for UC in East Asian populations. The G / A polymorphism of TNF-α-308 was not associated with UC in European populations.
