目的:通过检测miR-223、miR-210在糖尿病合并缺血性脑卒中患者血浆中的表达水平,探讨其与糖尿病合并缺血性脑卒中的关联。方法:采用病例对照研究,分单纯糖尿病、单纯缺血性脑卒中、糖尿病合并缺血性脑卒中及健康体检者4组,提取血浆总RNA,RT-PCR检测miR-223、miR-210的表达量,比较各组RNA的表达差异;分析miR-223、miR-210与缺血性脑卒中发生的关联性。结果:患者组miR-223、miR-210的表达水平均高于健康对照组(均P<0.05)。多因素Logistic回归分析显示,表达水平上调的miR-223、miR-210与缺血性脑卒中的发生有显著关联。结论:血浆miR-223、miR-210的表达水平与糖尿病合并缺血性脑卒中、单纯缺血性脑卒中的发生有显著关联,血浆miR-223、miR-210有望成为糖尿病合并缺血性脑卒中、单纯缺血性脑卒中的诊断指标。
OBJECTIVE: To detect the expression of miR-223 and miR-210 in the plasma of patients with diabetes mellitus complicated with ischemic stroke, and to explore its association with diabetes with ischemic stroke. Methods: A total of four groups of patients with simple diabetes mellitus, simple ischemic stroke, diabetes with ischemic stroke and healthy subjects were enrolled in this study. Plasma total RNA was extracted and the expression of miR-223 and miR-210 was detected by RT-PCR The expression of miR-223, miR-210 and ischemic stroke were analyzed. Results: The expression levels of miR-223 and miR-210 in patients were significantly higher than those in healthy controls (all P <0.05). Multivariate Logistic regression analysis showed that miR-223 and miR-210 were significantly associated with the occurrence of ischemic stroke. Conclusions: The expression of plasma miR-223 and miR-210 is significantly associated with the occurrence of ischemic stroke and ischemic stroke in diabetic patients. Plasma miR-223 and miR-210 are expected to become diabetic with ischemic brain Stroke, simple ischemic stroke diagnostic indicators.
