瞬时受体电位melastatin8(transient receptor potential melastatin8,TRPM8)蛋白是瞬时受体电位(transientreceptor potential,TRP)蛋白超家族中的一员,负责调控细胞内游离钙离子的动态平衡,在细胞增殖、凋亡以及分化中起着重要的作用。TRPM8最初作为一种前列腺特异性蛋白被克隆出来,之后发现其在肺癌、乳腺癌、结肠癌以及皮肤来源的恶性肿瘤中均有表达。最新研究发现,TRPM8不仅能感知冷知觉,还可作为前列腺癌诊断和治疗的新靶点。本文对TRPM8的结构、功能、定位、调控及其在前列腺癌发生和发展中的作用研究进行回顾,并对其在前列腺癌诊断和治疗中的应用前景进行展望。
Transient receptor potential melastatin8 (TRPM8) protein is a member of the superfamily of transient receptor potential (TRP) proteins and regulates the homeostasis of intracellular free calcium ion. TRPM8 was originally cloned as a prostate-specific protein and was subsequently found to be expressed in lung, breast, colon and skin-derived malignancies. The latest study found that TRPM8 can not only perceive cold perception, but also as a new target for the diagnosis and treatment of prostate cancer. In this review, we reviewed the study on the structure, function, localization, regulation and the role of TRPM8 in the pathogenesis and progression of prostate cancer, and prospected its application in the diagnosis and treatment of prostate cancer.
