应用冷冻蚀刻电镜技术,我们观察到小鼠正常胃粘膜上皮细胞膜的间隙连接和紧密连接。人胃癌MGC-803细胞膜上未见到间隙连接和紧密连接,表明胃癌细胞间隙连接结构的消失是细胞连接通讯抑制的主要原因。促癌变剂TPA和黄芫花分别处理胃粘膜导致膜上皮细胞的间隙连接消失或明显减少。本结果支持关于细胞连接通讯抑制是胃癌癌变机理之一,可能与癌变的促进有关的分析。本文对细胞间隙连接数目变化快的现象及可能的调节机理进行讨论。
Using cryo-etching electron microscopy, we observed gap junction and tight junction in the mouse normal gastric epithelial cell membrane. No gap junctions and tight junctions were found on the cell membrane of human gastric cancer MGC-803 cells, indicating that the disappearance of the gap junctional structure of gastric cancer cells is the main reason for inhibition of cell junctions. Carcinogen TPA and Daphne genkwa treatment of gastric mucosa respectively, leading to the disappearance of the gap junction membrane epithelial cells or significantly reduced. This result supports the observation that inhibition of cell-junctional communication is one of the mechanisms of carcinogenesis of gastric cancer and may be related to the promotion of carcinogenesis. This article discusses the phenomenon of rapid changes in the number of cell gap junctions and possible regulatory mechanisms.
