目的:观察酸味中药复方对糖尿病大鼠肾脏糖基化终产物(advanced glycation end products,AGEs)含量及其受体(receptor for advanced glycation end products,RAGE)mRNA表达的影响,探讨其防治糖尿病大鼠肾脏病变作用机制。方法:采用腹腔注射链脲佐菌素(streptozotoc in,STZ)配合高脂高热量饲料喂养复制糖尿病模型,采用荧光法、实时荧光定量PCR法分别检测糖尿病大鼠肾脏组织AGEs含量及RAGEmRNA表达。结果:糖尿病大鼠AGEs含量及RAGEmRNA表达明显高于正常大鼠(P<0.05);中药组及氨基胍组AGEs含量及RAGEmRNA表达明显低于糖尿病大鼠(P<0.05)。结论:酸味中药复方可能通过抑制AGEs的形成及RAGEmRNA表达实现其保护糖尿病大鼠肾脏作用的。
OBJECTIVE: To observe the effects of sour herb compound prescription on the level of diabetic glycation end products (AGEs) and their receptors for advanced glycation end products (RAGE) mRNA expression in diabetic rats, and to explore the prevention and treatment of diabetic rats Kidney disease mechanism. METHODS: Diabetic models were induced by intraperitoneal injection of streptozotocin in streptozotocin (STZ) combined with high-fat and high-calorie diets. The levels of AGEs and RAGE mRNA expression in kidneys of diabetic rats were detected by fluorescence and real-time fluorescence quantitative PCR. Results: The content of AGEs and the expression of RAGE mRNA in diabetic rats were significantly higher than those in normal rats (P<0.05). The contents of AGEs and RAGE mRNA expression in Chinese medicine group and aminoguanidine group were significantly lower than those in diabetic rats (P<0.05). Conclusion: The Chinese herbal compound with sour taste may achieve its role in protecting the kidney of diabetic rats by inhibiting the formation of AGEs and expression of RAGE mRNA.
