为探讨镉对睾丸结构和功能损害及锌对其保护作用的分子机制 ,研究镉对生精细胞凋亡的影响和锌的保护作用 ,选用 2月龄 Wistar雄性大鼠 2 4只 ,均分为染镉组、镉加锌组和对照组 ,分别自腹腔注射氯化镉 (2 mg/ kg BW) ,注射氯化镉同时及其前后 2 h各注射醋酸锌一次和等量生理盐水 ,7d后采用原位缺口平移技术 (INST)检测各组睾丸生精细胞凋亡数目的变化。结果显示与对照组相比 ,染镉组大鼠睾丸生精细胞凋亡数目明显增多 (P<0 .0 1,t=3.87) ;镉加锌组睾丸生精细胞凋亡数目无明显变化 (P>0 .0 5 ,t=1.34 )。染镉后大鼠睾丸生精细胞凋亡明显增加 ,锌对镉所诱导的生精细胞凋亡有保护作用 ,这些变化可能是镉损害睾丸结构和功能以及锌对其起保护作用的分子机制之一。
To explore the molecular mechanism of cadmium on testicular structure and function damage and zinc’s protective effect, to study the effect of cadmium on apoptosis of spermatogenic cells and the protective effect of zinc, 24 male Wistar rats of 2 months old were selected and divided equally into Cadmium group, cadmium plus zinc group and control group were injected intraperitoneally with cadmium chloride (2 mg / kg BW), cadmium chloride injection at the same time and before and after 2 h each injection of zinc acetate and an equal amount of saline, 7d In situ nicking shift (INST) was used to detect the changes of spermatogenic cell apoptosis in each group. The results showed that compared with the control group, the number of apoptotic spermatogenic cells in testis significantly increased (P <0.01, t = 3.87); there was no significant change in the number of apoptotic spermatogenic cells P> 0 .0 5, t = 1.34). Cadmium significantly increased the apoptosis of spermatogenic cells in testis of rats, and zinc could protect the apoptosis of spermatogenic cells induced by cadmium. These changes may be the molecular mechanism by which cadmium damage the structure and function of testis and the protective effect of zinc on it one.
