为探索生理范围内的静水压对体外培养的人膀胱平滑肌细胞(HBSMCs)的影响,实验采用自行设计的静水压加压装置并按压力梯度分组,用HE染色观察细胞形态,用免疫荧光、流式细胞仪、逆转录聚合酶链式反应(RT-PCR)分别检测细胞骨架F-actin、细胞周期、增殖细胞核抗原(PCNA)与基质金属蛋白酶7(MMP-7)。发现各组的细胞形态、骨架、细胞周期和增殖无明显差异,而各组间MMP-7mRNA的表达呈现出随静水压力的升高而增加随后逐渐下降的趋势。因此,我们得出生理范围内持续静水压可能对体外培养的HBSMCs的形态、骨架、周期、增殖无明显影响,而对MMP-7mRNA的表达具有一定的影响。
In order to explore the effect of hydrostatic pressure in physiological range on cultured human bladder smooth muscle cells (HBSMCs) in vitro, the hydrostatic pressure device designed by ourselves was used and divided into groups according to the pressure gradient. HE staining was used to observe the cell morphology. Immunofluorescence , F-actin, cell cycle, proliferating cell nuclear antigen (PCNA) and matrix metalloproteinase 7 (MMP-7) were detected by flow cytometry and reverse transcription polymerase chain reaction (RT-PCR). The results showed that there was no significant difference in the cell morphology, cytoskeleton, cell cycle and proliferation among groups. However, the expression of MMP-7mRNA increased with the increase of hydrostatic pressure and then gradually decreased. Therefore, we conclude that sustained hydrostatic pressure in the physiological range may have no significant effect on the morphology, skeleton, cycle and proliferation of HBSMCs cultured in vitro, but have a certain impact on the expression of MMP-7 mRNA.
