目的研究聚乳酸枝联CD40单克隆抗体在树突状细胞(DC)培养中的作用,从而对三维立体培养有深入的认识,为体外扩增树突状细胞提供新思路。方法利用鼠抗人CD40单克隆抗体对DC的诱导效应,观察聚乳酸枝联CD40单克隆抗体诱导的树突状细胞数量及生物学功能变化。结果与对照组普通悬液培养体系树突状细胞数量(5.43±33.31)×10~8比较,实验组三维立体培养系统树突状细胞数量(8.22±25.25)×10~8扩增(P<0.05)。实验组三维立体培养系统在6、9 d促进人树突状细胞分泌白细胞介素-12b(IL-12b)量高于对照组(P<0.05)。与对照组比较,实验组更加能促进人树突状细胞的成熟,促进其表面共刺激分子表达水平上调。结论通过CD40单克隆抗体与聚乳酸支架枝联建立的三维立体培养系统,有利于树突状细胞数量的扩增及其功能的增进。
Objective To study the role of polyclonal antibody against CD40 in the dendritic cell (DC) culture and to gain a better understanding of the three-dimensional culture and to provide new ideas for the dendritic cells to proliferate in vitro. Methods The induction of DCs by murine anti-human CD40 monoclonal antibody was used to observe the changes of dendritic cells and biological functions induced by polyclonal antibody against CD40. Results Compared with the control group, the number of dendritic cells (5.43 ± 33.31) × 10 ~ 8 in three-dimensional culture system of normal suspension increased (8.22 ± 25.25) × 10-8 (P < 0.05). The amount of interleukin-12b (IL-12b) secreted by dendritic cells in experimental group was significantly higher than that in control group at 6 and 9 days (P <0.05). Compared with the control group, the experimental group can further promote the maturation of human dendritic cells and promote the expression of co-stimulatory molecules on the surface. Conclusion The three-dimensional culture system established by CD40 monoclonal antibody and poly (DL-lactic acid) scaffolds is favorable for the expansion of the number of dendritic cells and the enhancement of its function.
