[目的]探讨翻白草丹参合剂对2型糖尿病(T2DM)大鼠模型过氧化物酶增殖受体γ(PPAR-γ)基因表达作用的分子机制.[方法]用小剂量链脲佐菌素(STZ)加高糖高脂饲料喂养大鼠制作T2DM大鼠模型.将大鼠随机分为正常对照组、模型对照组、阳性对照组和药物治疗组,分别灌胃给予蒸馏水、马来酸罗格列酮、翻白草丹参合剂12周.检测大鼠体质量、空腹血糖(FBC),总胆固醇(TG)、三酰甘油(TC)、高密度脂蛋白胆固醇(HDL-C)及低密度脂蛋白胆固醇(LDL-C)水平.并采用PT-PCR方法检测大鼠大网膜PPAR-γ基因表达.[结果]与正常对照组比较,模型对照组体质量显著降低(P<0.001),FBC、TC、TG、LDL-C值显著增高(P<0.001);与模型对照组比较,阳性对照组和药物治疗组体质量显著增高(P<0.05),FBC、TC、TG值显著降低(P<0.05),PPAR-γmRNA表达水平显著上调(P<0.05).[结论]翻白草丹参合剂可能通过提高PPAR-γ基因表达参与降低血糖及改善脂代谢紊乱.
[Objective] To investigate the molecular mechanism of the effect of Radix et Rhizoma Bletillae Salviae miltiorrhizae on gene expression of peroxisome proliferator-receptor gamma (PPAR-γ) in type 2 diabetes mellitus (T2DM) rats. [Methods] (STZ) plus high-fat and high-fat diet to make T2DM rat model.All the rats were randomly divided into normal control group, model control group, positive control group and drug treatment group, respectively, by intragastric administration of distilled water, maleate Luo The contents of body weight, fasting blood glucose (FBC), total cholesterol (TG), triglyceride (TC), high density lipoprotein cholesterol (HDL-C) and low density (P <0.001). Compared with the normal control group, the body mass of the model control group was significantly lower (P <0.001), and the expression of PPAR-γ in the omental tissue was detected by PT- FBC, TC, TG and LDL-C were significantly increased (P <0.001). Compared with the model control group, the body weight of the positive control group and the drug treatment group were significantly increased (P <0.05) P <0.05), PPAR-γmRNA expression was significantly up-regulated (P <0.05). [Conclusion] The treatment of Radix Salviae Miltiorrhizae could increase the expression of PPAR- Improve blood sugar and lipid metabolism disorders.
