目的观察川西獐牙菜75%乙醇提取物(SME)的镇痛作用和对咖啡因依赖小鼠戒断反应的抑制作用。方法将小鼠随机分为4组,分别以生理盐水、咖啡因和SME不同组合处理小鼠,每天两次,连续10 d;第10天初次处理后,注射纳洛酮引起戒断综合症,观察SME对戒断反应的抑制作用;在实验的第1、3、6、9、10天,用热板实验观察SME的镇痛作用;将小鼠随机分为空白组、模型组、阳性(阿司匹林)组和SME不同剂量组,分别给予相应药物,qd,连续8 d,通过扭体实验和血清生化指标的检测,进一步验证SME的镇痛作用。结果 SME可抑制咖啡因引起的镇痛高峰期,减少小鼠在戒断反应中的跳跃次数;扭体实验中,给予小鼠不同剂量SME后,扭体次数和给药剂量之间呈负相关,并明显降低血清中前列素E2和一氧化氮的含量。结论 SME有显著的镇痛作用和潜在的抑制戒断反应的作用。
Objective To observe the analgesic effect of Swertia chinensis 75% ethanol extract (SME) and its inhibitory effect on withdrawal response in caffeine-dependent mice. Methods Mice were randomly divided into 4 groups. The mice were treated with different combinations of saline, caffeine and SME respectively, twice a day for 10 days. After the first treatment on day 10, the injection of naloxone caused withdrawal syndrome, The inhibitory effect of SME on withdrawal response was observed. On the 1st, 3rd, 6th, 9th and 10th day of the experiment, the analgesic effect of SME was observed by hot plate test. The mice were randomly divided into blank group, model group, Aspirin) and SME different dose groups were given the corresponding drugs, qd, 8 consecutive days, by writhing test and serum biochemical indicators to further verify the analgesic effect of SME. Results SME could inhibit the peak of analgesia induced by caffeine and reduce the number of jumps in mice during withdrawal response. In the writhing experiment, the number of writhing was negatively correlated with the dose of SME administered to mice , And significantly reduced serum levels of prostaglandin E2 and nitric oxide. Conclusion SME has significant analgesic effect and potential inhibition of withdrawal response.
