本文介绍用~3H-TdR掺入示踪技术,探讨白细胞介素2(IL-2)对辐射引起人血淋巴细胞增殖抑制的逆转作用。用~(60)Coγ射线体外照射人血淋巴细胞后,加入植物血球凝集素(PHA)和外源性IL-2常规培养,以淋巴细胞DNA中~3H-TdR掺入量的变化来衡量T淋巴细胞的增殖能力。结果表明,1-40Gy剂量的γ射线照射均可引起T淋巴细胞增殖力抑制,40Gy剂量组增殖率只有27.6%;加IL-2后,1-10Gy剂量组的T淋巴细胞增殖幸均明显增加,提示IL-2有使受辐射损伤的T淋巴细胞增殖能力恢复的效应。
This article describes the use of ~3H-TdR incorporation of tracer technology to investigate the reversal effect of interleukin 2 (IL-2) on inhibition of proliferation-induced human blood lymphocyte proliferation. After in vitro irradiation of human blood lymphocytes with ~(60)Co γ-rays, phytohemagglutinin (PHA) and exogenous IL-2 were routinely cultured to measure the incorporation of ~3H-TdR in lymphocyte DNA. Lymphocyte proliferation. The results showed that 1-40Gy dose of γ-ray irradiation can cause T lymphocyte proliferation inhibition, 40Gy dose group proliferation rate was only 27.6%; plus IL-2, 1-10Gy dose group T lymphocyte proliferation were significantly increased It suggests that IL-2 has the effect of restoring the proliferation ability of radiation-damaged T lymphocytes.
