通过对缺铁性贫血(IDA)大鼠红细胞膜带3蛋白的研究,探讨缺铁对红细胞膜蛋白的影响及其在IDA发病机理中的作用。实验表明,铁缺乏可引起红细胞膜蛋白发生异常变化:带3蛋白与带4.1蛋白的相对含量比值下降甚至可完全倒置,收缩蛋白减少;带3蛋白的阴离子(Cl~-)交换功能减弱。膜蛋白相对含量变化出现的快慢和强度与缺铁严重程度和缺铁病程有关。结果提示,(1)带3等膜蛋白的缺损可能发生在骨髓红细胞合成期,这与IDA红细胞体积减小密切有关;(2)铁缺乏对红细胞膜蛋白影响的最终结果是破坏膜骨架的结构与功能,使红细胞变形能力下降,易被破坏吞噬。这可能是IDA红细胞寿命缩短的主要原因。
To investigate the effect of iron deficiency on erythrocyte membrane protein and its role in the pathogenesis of IDA by studying erythrocyte membrane protein 3 in rats with iron deficiency anemia (IDA). Experiments show that iron deficiency can cause abnormal changes in membrane proteins of erythrocytes: the ratio of the relative content of band 3 protein and band 4.1 protein is reduced or even completely inverted, and the number of contractile proteins is reduced; the anion exchange function of band 3 protein is weakened. The relative content of membrane protein changes in the speed and intensity of iron deficiency severity and iron deficiency related to the course. The results suggest that (1) the defect of membrane protein with 3 may occur in the stage of erythrocyte synthesis, which is closely related to the decrease of red cell volume of IDA. (2) The final result of the effect of iron deficiency on erythrocyte membrane protein is to destroy the structure of membrane skeleton And function, so that the ability to deform erythrocytes, easily destroyed swallowed. This may be the main reason for the shortened lifespan of IDA red blood cells.
