目的 探讨光动力疗法 (PDT)对人鼻咽癌细胞株HNE1凋亡的作用。方法 采用从光导纤维输出的氦氖激光 ,功率 15 0mW ,波长 6 32 .8nm ,能量密度 10J/cm2 ,照射含癌光啉光敏剂的鼻咽癌细胞 ,分别在PDT后 3,6 ,12 ,2 4,36和 48h后 ,光镜和电镜观察凋亡细胞的形态学改变 ,并计数细胞凋亡率 (% )。结果 人鼻咽癌细胞株HNE1经PDT 2 4h后出现细胞凋亡的典型形态学改变 ,12h后出现早期凋亡细胞的形态特征 ,光镜和电镜下可见细胞大小不一 ,细胞质浓缩 ,核固缩 ,染色质边集 ,进而细胞裂解和凋亡小体形成。细胞凋亡率分别为 0 % ,1% ,6 % ,71% ,86 %和 95 %。细胞超微结构改变的严重程度依PDT后时间的增加而逐渐递增。结论 PDT对HNE1细胞具有明显杀伤效应 ,并诱导其发生凋亡
Objective To investigate the effect of photodynamic therapy (PDT) on the apoptosis of human nasopharyngeal carcinoma cell line HNE1. Methods The nasopharyngeal carcinoma cells with photopheresis photosensitizer were irradiated with He-Ne laser output from optical fiber at a power of 15 0 mW, a wavelength of 6 32 .8 nm and an energy density of 10 J / cm2. The rates of apoptosis were 0%, 1%, 6%, 71%, 86% and 95%, respectively. The severity of cell ultrastructural changes gradually increased with the increase of PDT time. Conclusion PDT has obvious killing effect on HNE1 cells and induces apoptosis of HNE1 cells
