蜥蜴胃康基本方及其拆方对胃癌前病变模型大鼠HIF-1α、VEGF表达影响的实验研究

目的研究蜥蜴胃康基本方对胃癌前病变(PLGC)模型大鼠缺氧诱导因子HIF-1α和血管内皮生长因子VEGF表达的影响,探讨蜥蜴胃康基本方对PLGC的调控机制。方法将70只SD雄性大鼠随机分成空白对照组、胃癌前病变模型组、蜥蜴胃康基本方全方组、补气组、养阴组、解毒通络组、维酶素对照组。除空白对照组外,余组均采用浓度0.02mmol/L的N-甲基-N’-硝基-N-亚硝基胍(MNNG)灌胃及自由饮用、40%乙醇灌胃、饥饱失常综合因素,共24周来制备胃癌前病变大鼠模型。分别用蜥蜴胃康基本方、补气药、解毒通络药、养阴药、维酶素连续治疗12周,观察大鼠胃黏膜病理组织学变化,应用免疫组化法检测胃体组织HIF-1α和VEGF因子表达情况。结果通过Image-Pro Plus6.0图像分析系统对免疫组化图像进行光密度的测算,各组与模型组比较P<0.05;各治疗组与空白对照组比较P<0.05;蜥蜴胃康基本方全方及补气、解毒通络治疗组HIF-1α、VEGF因子阳性表达率低于维酶素治疗组(P<0.05),其中以全方组AOD值最小;三组拆方比较P<0.05,其中补气组AOD值最小,解毒通络组次之,养阴组表达较高。结论蜥蜴胃康基本方及其拆方能部分逆转PLGC模型大鼠胃黏膜病理变化,降低HIF-1α、VEGF阳性表达,可促进细胞凋亡,抑制细胞过度增殖,这可能是蜥蜴胃康基本方干预PLGC的分子生物学机制。 Objective To study the effect of likankie kang basic recipe on hypoxia inducible factor (HIF-1α) and vascular endothelial growth factor (VEGF) expression in gastric precancerous lesions (PLGC) model rats, and to explore the regulation mechanism of lizards weikang basic formula on PLGC. The AOD values ​​of all the prescriptions were the lowest. The three prescriptions were significantly lower than P <0.05, and the positive rates of HIF-1α and VEGF were lower Among them, the AOD value of qi supplement group was the lowest, followed by that of detoxification and mending group, while that of yin-yin group was the highest. Conclusion Lizhenweikang Decoction can partly reverse the pathological changes of gastric mucosa in rats with PLGC and decrease the positive expression of HIF-1α and VEGF, which can promote cell apoptosis and inhibit the proliferation of cells, which may be the basic form of Lizo Weikang Intervention in molecular biology of PLGC.
