目的 :研究Ku70基因导入人肺癌细胞 (LCA)后的表达及意义。方法 :构建重组的正义和反义Ku70基因表达质粒载体分别导入LCA细胞。结果 :用Northern印迹杂交法证明Ku70正义、反义基因均可在LCA的mRNA中得到表达 ,用West ern blotting法去证明细胞中只有导入正义Ku70基因的蛋白表达 ,而导入反义Ku70基因的人肺癌细胞 ,无正义Ku70蛋白表达 ,结论 :正义Ku70基因的表达可以调节DNA的修复 ,反义Ku70基因的表达可以增强肿瘤细胞对放射线照射的敏感性。这些表达特性可为我们治疗肿瘤提供新的分子生物学手段
Objective: To study the expression and significance of Ku70 gene in human lung cancer cells (LCA). METHODS: Construction of recombinant plasmids expressing the sense and antisense Ku70 gene were introduced into LCA cells, respectively. RESULTS: Northern blot hybridization was used to verify that the Ku70 sense and antisense genes were expressed in the LCA mRNA. Western blotting was used to prove that only the protein expressing the Ku70 gene was introduced into the cell, whereas the antisense Ku70 gene was introduced into the cell. In lung cancer cells, there was no sense of Ku70 protein expression. Conclusion: The expression of just Ku70 gene can regulate DNA repair, and the expression of antisense Ku70 gene can enhance the sensitivity of tumor cells to radiation irradiation. These expression characteristics can provide us with new molecular biological methods for treating tumors
