本实验运用受体放射自显影术观察神经毒素甲基-苯基-四氢吡啶(MPTP)在大鼠脑内的分布,并运用亚细胞成份分离技术研究MPTP在小鼠脑亚细胞成分中的分布。结果表明,~3H—MPTP在大鼠脑内有其特异性的结合位点,这一位点能被非标记的MPTP和单胺氧化酶抑制剂优降宁竞争结合。~3H-MPTP亚细胞的分布显示,该物质与线粒体的结合量最大。以上结果提示,~3H—MPTP在脑内的受体可能为单胺氧化酶,该酶在帕金森病的发病机理中有可能起着某种重要的作用。
In this study, we used receptor autoradiography to observe the distribution of neurotoxin methyl-phenyl-tetrahydropyridine (MPTP) in the rat brain. We used subcellular fractionation to study the effect of MPTP on the brain subcellular components of mice distributed. The results showed that ~ 3H-MPTP had its specific binding site in rat brain, which could be competitively bound by unlabeled MPTP and monoamine oxidase inhibitor biniban. ~ 3H-MPTP subcellular distribution showed that the substance and mitochondria the largest amount of binding. These results suggest that ~ 3H-MPTP receptors in the brain may be monoamine oxidase, the enzyme in the pathogenesis of Parkinson’s disease may play an important role.
